SAF transport medium broth is used to preserve fecal specimen for all intestinal parasites at room temperature for later examination : Exam by wet mount, concentration and permanent stain procedures.

STORAGE
Store prepared media at 25º C protected from direct light.

PROCEDURE FOR SPECIMEN COLLECTION
Collect specimens as previously standardized, following collection procedures. Kit systems are packaged in Biobags with illustrated multi-lingual instructions to assist in safe and sanitary specimen collection by personnel and/or patients. Bring to the laboratory as soon as possible.


PROCEDURE

Wet Mount Examination

  1. On a slide, place a drop of SAF-stool mixture and a drop of saline/iodine.
  2. Overlay with a coverglass.
  3. Examine microscopically for presence of parasites.

Permanent Smears

  1. On a slide, place a drop of SAF-stool mixture and a drop of Mayer's Albumine (8951).
  2. Spread to cover slide and to form a thin film.
  3. Let slides to dry at room temperature.
  4. Fix the film into a 70% ethanol solution bath for 30 minutes
  5. Referring to parasites procedures, stain slides
  6. Examine microscopically for presence of parasites.

Concentration Procedure
Follow directions as described in Para-Test Kit (cat. No. PT-50 or PT-50V).

LIMITATIONS OF METHOD
Inadequate fixation or too little/too much specimen may give false results. This is only a part of the identification. Others tests may be required.
